I have my iTunes media, currently about 400 gigs of music data, stored on a pair of 2tb external drives, one the primary, the second a backup (JBOD, not RAID). At what point is it recommended to run the defragment utility on these disks? Thanks for your suggestions.

I would start by asking Windows to analyze the drives in the Disk Manager. If Windows reports the drive fragmented then it's good to defragment. I used to run an automated defragmenting script nightly on an old machine of mine :~)
